Detection of Ureaplasma urealyticum from Amniotic Fluid using Conventional Method and Polymerase Chain Reaction

Abstract
The involvement of Ureaplasma urealyticum in various pregnancy complications has been previously known, especially in chorioamninonitis, premature rupture of membranes, recurrent abortions and perinatal morbidity and mortality. To isolation and
detection of U.urealyticum, We can use conventional culture method and Polymerase Chain Reaction, There are two kind of culture method, one is broth culture and the other is agar culture. The broth culture is more convenient than agar culture
method. In
this study, we compared broth culture method and PCR. We used urea broth for culture method and primer for PCR is proposed by J.J Willougby. We incubated urea broth for 7days. When change the broth to pink color, we decided positive result. But
no
color
change after 7 days, decieded negative result. PCR condition was 94¡É/1min, 72¡É/1min, 55¡É/1min, and repeated 35 times.
@ES We obtained the results as follows.
@EN 1. The isolation rate tf U.urealyticum in culture method was 12 cases (24%), but 14% cases (28%)in PCR.
2. Elapsed times to obtain positive results in culture method showed 2 cases after 24 hours. 4 cases after 48 hours, 3 cases after 72 hours, 2 cases after 96 hours and 1 case after 120 hours.
3. We obtained the conclusion as PCR is more sensitive than conventional culture method using urea broth.
